Effects of Repeated Electroconvulsive Shock on [3H]MK-801 Binding in Rat Brain



Abstract
Electroconvulsive shock(ECS) is an effective treatment modality to various psychiatric disorders, but we do not have a detailed and complete understanding of its mechanisms of action, In order to elucidate the therapeutic mechanism of ECS in the
schizophrenia, the authors measured the maximal binding capacity(Bmax) and dissociation constant(Kd) of the binding sites of [3H]MK-801, which might be related with the pathophysiology of schizophrenia, using rat brains.
We administered seven daily ECS to the experimental group(n=8), and the same procedure with sham-ECS to the control group(n=8)
There was no significant difference between two groups in the number and the affinity of the[3H]MK-801 binding sites. The results suggested that the mechanisms of action of ECS might not be worked through the PCP receptor.
However, it should be replicated with larger samples and more controlled experimental design gecause this kind of research has not been performed until this study. And we think that further studies are needed including other subtypes of
excitatory
amino
acid receptors.
